A reaction of the above-mentioned N,N-dialkyl-N’-chlorosulfonyl chloroformamidine derivatives with N1-substituted 5-pyrazolones afforded pyrazolo[4,3-e][1,4,3]oxathiazine derivatives by a selective 1,3-CCO dinucleophilic substitution. [1,2,4]Triazolo[1,2-b][1,2,3,5]thiatriazole derivatives and pyrazolo[4,3-e][1,4,3]oxathiazine derivatives were the only products isolated from the resp. reactions and both represent new ring systems.

A novel synthesis of trimethylsilyldiazomethane (TMSCHN2) by diazotization of trimethylsilylmethylamine (TMSCH2NH2) is reported using batch and continuous flow syntheses. The latter affords a daily production of 275 g (2.4 mol) of TMSCHN2. Other silylated methylamines were also successfully reacted under the developed reaction conditions to furnish various silicon-bearing diazomethane reagents. The applicability of the process is highlighted by disclosure of batch and continuous flow one-pot esterification and 1,3-dipolar cycloaddition processes. Furthermore, the high-yielding esterification of carboxylic acids with silylated and substituted methylamines in continuous flow is disclosed. Finally, work-up and purification procedures are reported for the preparation of a 2-MeTHF solution of TMSCHN2, which can be used in rhodium-catalyzed methylenation and homologation reactions.

High-throughput screening for the induction of a luciferase reporter gene in a thrombopoietin (TPO)-responsive cell line resulted in the identification of 4-diazo-3-hydroxy-1-naphthalenesulfonic acids as TPO mimics. Modification of the core structure and adjustment of unwanted functionality resulted in the development of (5-oxo-1,5-dihydropyrazol-4-ylidene)hydrazines which exhibited efficacies equivalent to those of TPO in several cell-based assays designed to measure thrombopoietic activity. Furthermore, these compounds elicited biochem. responses in TPO-receptor-expressing cells similar to those in TPO itself, including kinase activation and protein phosphorylation. Potencies for the best compounds were high for such low mol. weight compounds (MW < 500) with EC50 values in the region of 1-20 nM.

In the preparation of the title dyes, an aryldiazonium chloride is coupled in alk. medium with 1-phenyl-3-methyl-5-pyrazolone or 1-(2-chlorophenyl)-3-methyl-5-pyrazolone and the second arylazo group in position-4 of the 1-Ph group is introduced in acidic medium and the third arylazo group in position-7 of the 4-naphthyl group of H- or γ-acid is applied in alk. medium. Light fastness on wool was 4-5 to 5-6 and 3-4 to 5-6 on polyamide. Wash fastness was 3-4 to 4-5 on wool and 2-3 to 3-4 on polyamide.
