This resulted in the discovery of \u03b1,\u03b1,\u03b1-trifluorotolyl I which exhibits 800 x selectivity for \u03b1v\u03b25vs. \u03b1v\u03b23 with a pyrrolidine amide linker that confers selectivity for \u03b1v\u03b25 by positioning a key aryl ring in the SDL of \u03b1v\u03b25 with good complementarity; binding in this mode is disfavored in \u03b1v\u03b23 due to clashes with key residues in the \u03b23-subunit.
