An updated downstream synthesis route of 143426-52-2 as follows.<\/p>\n<p>To a stirred solution of intermediate 1 (0.15 g, 0.73 mmol) in MeCN (10 mL), were added Intermediate 11 (0.14 g , 0.75 mmol) and trimethylamine (0.27 mL, 1.86 mmol ) at RT and the resulting mixture was stirred overnight at RT. After completion of reaction, the reaction mixture was quenched with water (10 mL) and extracted with EtOAc (3 x 30 ml_). The combined organic layer was washed with brine (30 mL), dried over Na2S04, filtered and concentrated under vacuum. The resulting crude material was purified by prep-HPLC (Method B) to get desired compound. Yield: 39% (90 mg, White solid). 1H NMR (400 MHz, DMSO-d6): d 8.45 (d, J = 2.0 Hz, 1 H), 7.77-7.73 (m, 3H), 7.35 (d, J = 8.4 Hz, 2H), 6.93-6.82 (m, 3H), 6.53 (s, 1 H), 4.22 (s, 4H), 3.72 (d, J = 13.2 Hz, 1 H), 3.33-3.27 (m, 1 H), 3.09-3.06 (m, 1 H), 2.97 (t, J = 7.6 Hz, 1 H), 2.19-2.06 (m, 2H), 1.81-1.71 (m, 2H), 1.70-1.56 (m, 1 H). LCMS: (Method A) 362.1 (M+H), Rt. 97.70 min, 88.96% (Max).
