Example 2 6-(4-(5-chloro-2-(1,5-dimethyl-1H-pyrazol-3-yl)amino)pyrimidin-4-ylamino)piperidin-1-yl)nicotinonitrile To a solution of 6-(4-((2,5-dichloropyrimidin-4-yl)amino)piperidin-1-yl)nicotinonitrile (100 mg, 0.286 mmol) in n-butanol (5 mL) was added 1,5-dimethyl-1H-pyrazol-3-amine (95 mg, 0.859 mmol) and TFA (212 muL, 2.86 mmol). The reaction mixture was stirred at 120 C. in a sealed tube for 24 hours then cooled to room temperature and diluted with EtOAc (50 mL), washed with water (15 mL*3) followed by brine (20 mL). The organic layer was dried over anhydrous Na2SO4, filtered and concentrated in vacuo. The residue was purified by silica gel column chromatography (DCM\/EA\/MeOH (v\/v\/v)=25\/5\/1) to give the title compound as a white solid (53 mg, yield 44%). LC-MS (ESI, pos. ion) m\/z: 423.7 [M+H]+; 1H NMR (300 MHz, DMSO-d6) delta (ppm): 9.17 (s, 1H), 8.49 (d, J=2.3 Hz, 1H), 7.87 (s, 1H), 7.84 (dd, J=9.1, 2.3 Hz, 1H), 7.00 (d, J=9.2 Hz, 1H), 6.78 (d, J=7.8 Hz, 1H), 6.35 (s, 1H), 4.57-4.51 (m, 2H), 4.32-4.19 (m, 1H), 3.60 (s, 3H), 3.07-2.99 (m, 2H), 2.22 (s, 3H), 2.00-1.95 (m, 2H), 1.65-1.52 (m, 2H).<\/p>\n
