A combined screening strategy using HTS together with focused kinase library and virtual screening led to the identification of diverse chem. series as PDK1 inhibitors. The authors focused the medicinal chem. efforts on 7-azaindoles with low micromolar IC50s, e.g. I (IC50 = 1.1 μM) in the biochem. PDK1 assay. The structure-guided optimization efforts considered also PDK1 x-ray structures with weaker binding fragments and resulted in 7-azaindoles with significantly improved biochem. PDK1 potency in the two-digit nanomolar range. However, the most potent analogs only showed moderate activities in a cellular mechanistic assay, e.g. II (IC50 = 2.3 μM) together with either low microsomal stability or low permeability. The described structure-activity relationship together with PDK1 x-ray structures and early ADME data provided the basis for the subsequent hit-to-lead program.

G-protein coupled receptor kinase 2 (GRK2), which is upregulated in the failing heart, appears to play a critical role in heart failure (HF) progression in part because enhanced GRK2 activity promotes dysfunction of β-adrenergic signaling and myocyte death. An orally bioavailable GRK2 inhibitor could offer unique therapeutic outcomes that cannot be attained by current heart failure treatments that directly target GPCRs or angiotensin-converting enzyme. Herein, we describe the discovery of a potent, selective, and orally bioavailable GRK2 inhibitor, 8h, through high-throughput screening, hit-to-lead optimization, structure-based design, mol. modeling, synthesis, and biol. evaluation. In the cellular target engagement assays, 8h enhances isoproterenol-mediated cyclic adenosine 3′,5′-monophosphate (cAMP) production in HEK293 cells overexpressing GRK2. Compound 8h was further evaluated in a human stem cell-derived cardiomyocyte (HSC-CM) contractility assay and potentiated isoproterenol-induced beating rate in HSC-CMs.

Xu, Jiaojiao;Cai, Jin;Chen, Junding;Zong, Xi;Wu, Xuan;Ji, Min;Wang, Peng research published 《 An efficient synthesis of baricitinib》, the research content is summarized as follows. A highly efficient method for the synthesis of baricitinib was developed. The starting material tert-Bu 3-oxoazetidine-1-carboxylate was converted to intermediate 2-(1-(ethylsulfonyl)azetidin-3-ylidene)acetonitrile via the Horner-Emmons reaction, deprotection of the N-Boc-group and a final sulfonamidation reaction. Then the nucleophilic addition reaction was carried out smoothly to afford the borate intermediate in the presence of 1,8-diazabicyclo[5.4.0]undec-7-ene under reflux. Finally, the desired compound baricitinib was obtained by the Suzuki coupling reaction of 4-chloro-7-H-pyrrolo[2,3-d]pyrimidine with the above borate intermediate. All compounds were characterised by IR, MS, 1H NMR and 13C NMR. The overall yield in this synthetic route was as high as 49%. Moreover, this procedure was straightforward to carry out, had low cost and was suitable for industrial production

Herein, the design, synthesis, and structure-activity relationships of a series of unique 4-(1H-pyrazol-4-yl)-pyrimidin-2-amine derivatives that selectively inhibit Janus kinase 2 (JAK2) and FLT3 kinases is described. These screening cascades revealed that I was a preferred compound with IC50 values of 0.7 and 4 nM for JAK2 and FLT3, resp. Moreover, I was a potent JAK2 inhibitor with 37-fold and 56-fold selectivity over JAK1 and JAK3, resp., and possessed an excellent selectivity profile over the other 100 representative kinases. In a series of cytokine-stimulated cell-based assays, I exhibited a higher JAK2 selectivity over other JAK isoforms. The oral administration of 60 mg/kg of I could significantly inhibit tumor growth, with a tumor growth inhibition rate of 93 and 85% in MV4-11 and SET-2 xenograft models, resp. Addnl., I showed an excellent bioavailability (F = 58%), a suitable half-life time (T1/2 = 4.1 h), a satisfactory metabolic stability, and a weak CYP3A4 inhibitory activity, suggesting that I might be a potential drug candidate for JAK2-driven myeloproliferative neoplasms and FLT3-internal tandem duplication-driven acute myelogenous leukemia.

Yao, Lianbin;Ohlson, Sten;Dymock, Brian W. research published 《 Design and synthesis of triple inhibitors of janus kinase (JAK), histone deacetylase (HDAC) and Heat Shock Protein 90 (HSP90)》, the research content is summarized as follows. Inhibition of multiple signaling pathways in a cancer cell with a single mol. could result in better therapies that are simpler to administer. Efficacy may be achieved with reduced potency against individual targets if there is synergy through multiple pathway inhibition. To achieve this, it is necessary to be able to build multi-component ligands by joining together key pharmacophores in a way which maintains sufficient activity against the individual pathways. Designed triple inhibiting ligands are explored aiming to block three completely different target types: a kinase (JAK2), an epigenetic target (HDAC) and a chaperone (HSP90). Although these enzymes have totally different functions they are related through inter-dependent pathways in the developing cancer cell. Synthesis of several complex multi-inhibiting ligands are presented along with initial enzyme inhibition data against 3 biol. target classes of interest. A lead compound, 47 (I), was discovered which had low micromolar activity for all 3 targets. Further development of these complex trispecific designed multiple ligands could result in a ‘transient drug’, an alternative combination therapy for treating cancer mediated via a single mol.

Y.;Ohlson, Sten;Dymock, Brian W. research published 《 Design and Synthesis of Ligand Efficient Dual Inhibitors of Janus Kinase (JAK) and Histone Deacetylase (HDAC) Based on Ruxolitinib and Vorinostat》, the research content is summarized as follows. Concomitant inhibition of multiple oncogenic pathways is a desirable goal in cancer therapy. To achieve such an outcome with a single mol. would simplify treatment regimes. Herein the core features of ruxolitinib, a marketed JAK1/2 inhibitor, have been merged with the HDAC inhibitor vorinostat, leading to new mols. that are bispecific targeted JAK/HDAC inhibitors. A preferred pyrazole substituted pyrrolopyrimidine, I (R = H), inhibits JAK1 and HDACs 1, 2, 3, 6, and 10 with IC50 values of less than 20 nM, is <100 nM potent against JAK2 and HDAC11, and is selective for the JAK family against a panel of 97 kinases. Broad cellular antiproliferative potency of I (R = H) is supported by demonstration of JAK-STAT and HDAC pathway blockade in hematol. cell lines. Me analog I (R = Me) has an even more selective profile. This study provides new leads for assessment of JAK and HDAC pathway dual inhibition achieved with a single mol.

Although lung adenocarcinoma patients have benefited from the development of targeted therapy, patients with lung squamous cell carcinoma (SqCC) have no effective treatment due to the complexity and heterogeneity of the disease. Therefore, basing on the genetic anal. of mutations in lung squamous cell carcinoma to design multi-target inhibitors represents a potential strategy for the medical treatment. In this study, through screening an inhouse focused library, we identified an interesting indazole scaffold. And following with binding anal., we elaborated the structure-activity relationship of this hit compound by optimizing four parts guided by the DDR2 enzymic assay, which resulted in a potent lead compound 10a. We conducted further optimization of dual enzymic inhibitions towards FGFR1 and DDR2, two important kinases in lung squamous cell carcinoma. Finally, from the cellular antiproliferative activity tests and in vivo pharmacokinetic test, 3-substituted indazole derivative 11k was found to be a promising candidate and subjected to in vivo pharmacol. study with the mouse xenograft models, demonstrating profound anti-tumor efficacy. Addnl. in vitro druglike assessment reinforced that compound 11k could be valuable for SqCC drug development.

Vascular endothelial growth factor-2 (VEGFR-2) plays a pivotal role in tumor angiogenesis. Herein, a library of novel 2-(4-(1H-indazol-6-yl)-1H-pyrazol-1-yl)acetamide derivatives were designed and synthesized as VEGFR-2 inhibitors based on scaffold hopping strategy. These compounds exhibited excellent inhibitory activity in both VEGFR-2 and tumor cell proliferation. Especially, compound I possessed potent VEGFR-2 inhibition with IC50 = 1.6 nM and anti-proliferation against HGC-27 tumor cells with IC50 = 0.36 +/- 0.11μM, as well as less toxicity against normal GES-1 cells with IC50 = 187.46 +/- 10.13μM. Moreover, I obviously inhibited colony formation, migration and invasion of HGC-27 cells by adjusting the expression of MMP-9 and E-cadherin, and induced HGC-27 cells apoptosis by increasing ROS production and regulates the expression of apoptotic proteins. Furthermore, I blocked the PI3K-Akt-mTOR signaling pathway in HGC-27 cells. In addition, anti-angiogenesis of I was proved by inhibition tube formation and the expression of p-VEGFR-2 in HUVEC cells. All the results demonstrated that I could be developed as a promising anticancer agent for gastric cancer therapy.
